Adrian White, 42, and his 14-year-old son Harry, from St Brides Major, will set off on August 17 in tribute to wife and mum Claire White, who died in March at the Marie Curie Hospice, Cardiff and the Vale, aged 39.
The journey will include reaching the summit of Pen y Fan on August 20, which would have been Claire’s 40th birthday.
Mr White said: “We want to raise awareness of the incredible care that Marie Curie provides patients and their families during what was a very difficult and challenging time.”
Mrs White was admitted to the Penarth hospice in February following what her family described as a ‘brave fight with cancer.’
The pair described her as a ‘strong, gentle and beautiful person who was fiercely passionate about fighting for what is right.’
Mr White and Harry will begin their walk in Quedgeley, near Gloucester, and travel through the Forest of Dean, Monmouthshire, and Brecon before heading down the Taff Trail to Cardiff, then on to the hospice, ending at their family home in St Brides Major.

They plan to walk an average of 40km per day, carrying their equipment and camping along the way.
Mr White said: “This journey will provide Harry and I the perfect opportunity to reflect on our amazing life with Claire and continue to build our strong father and son bond.
“The walk is a form of therapy and also a bonding experience for us.”
They are aiming to raise £9,300 for Marie Curie – the cost of one day of care at the hospice.
